EPIGENETICS SERVICES SAMPLE SUBMISSION CHECK LIST

Please use this checklist to make sure your samples arrive safe and in a timely manner to our Services lab.

For International shipments, scientific logistic carriers such as World Courier are highly recommended. Customers may choose any shipping carriers however shipping is FCA (INCOTERMS 2020) Active Motif's facility, and Active Motif is not responsible for spoliation or other damage/loss during shipment.

Ensure that 5-10 kg (10-20 lbs.) of dry ice is included in shipment.
Secure samples in a box inside shipping container. Please avoid loose tubes in shipping container
Make sure name/number on sample tube matches the sample name/number listed on the Sample Submission form
Include quote number in Sample Submission form
Include completed Sample Submission form in shipment
Upon sample shipment, send an email notification containing the following information to episervices@activemotif.com (North America), euro-orders@activemotif.com (Europe), or japantech@activemotif.com (Japan):
- Quote number
- Shipment tracking number
- An electronic copy of sample submission form as attachment

IMPORTANT GUIDELINES:

Customer will prepare samples according to protocols provided by Active Motif.
We require customer samples to meet our quality standards before proceeding with the service. If the samples do not meet the required standards, we will notify the customer and may request replacement samples. Any additional processing required to assess the quality of the samples may be charged to the customer.
We cannot accept infectious material unless the sample is formaldehyde-fixed.
Samples must be shipped to Active Motif within 6 months of placing the purchase order. Delays in shipments to Active Motif may result in a delay to the project and price adjustments.
For projects including antibody sourcing, Active Motif will order the antibody upon receiving the purchase order and this portion of the project will be invoiced upon arrival of the antibody from the vendor.
For services on cryopreserved cells, please measure viability using Trypan Blue, AO/PI staining, flow cytometry, or other appropriate method before cryopreservation. Please thaw a test sample to perform your own viability check and indicate result in "Other Comments" section below.
For Single-Cell Services on cryopreserved cells, we measure viability and proceed with all samples that are >70% viable. Samples that are 50-70% viable are considered at-risk and we can proceed with your permission. Please indicate in the comments if you would like us to proceed with samples that are 50-70% viable. For samples <50% viable we will not proceed and we will ask for replacements.
Original samples and intermediate material, such as NGS libraries, will be stored at Active Motif for up to 12 months after data delivery. After 12 months, Active Motif reserves the right to discard the samples and intermediate materials. The return shipment of samples and/or libraries is available upon request and will be at the customer's expense for processing and shipping.
Data files will be available for 3 months after delivery, after which Active Motif reserves the right to delete the files. Active Motif is not responsible for data retention or retrieval after deletion, as specified in the Bioinformatics Analysis Request Form.

SHIPPING INSTRUCTIONS:

All samples pertaining to the order must be shipped together. Ship samples Monday through Wednesday. Do not ship over a weekend or for Saturday delivery.
Ship cell and tissue samples on 5-10 kg (10-20 lbs.) dry ice, overnight, for morning delivery using a suitable common carrier. Ensure that samples are contained in a box within the shipping container and not loose in shipping container. Purified DNA samples, including DNA libraries, can be shipped at -20°C.
Antibodies (if applicable) should be shipped overnight, for morning delivery using a suitable common carrier.

To ensure optimal data output and achieve the desired sequencing coverage, please adhere to the following minimum requirements for pooled libraries on the NovaSeq platform:

SP Flow Cells: 2 nM concentration, 40 µl volume
S1 Flow Cells: 2 nM concentration, 40 µl volume
S2 Flow Cells: 2 nM concentration, 50 µl volume
S4 Flow Cells: 2 nM concentration, 60 µl volume

For individual libraries, we recommend a concentration of 2 nM and a minimum volume of 15 µl per sample. Libraries prepared with a lower concentration or volume may result in reduced read counts for those samples. Additionally, a minimum Qubit quantification of 1 ng/µL is required to ensure sufficient DNA quality and yield.

To ensure accurate processing, please submit a separate form for each assay if you are submitting samples for multiple assays.

Please ensure that ALL required information is provided for each sample. If a field is not applicable, the information is unavailable, or cannot be disclosed due to confidentiality, enter “N/A”. If you are unsure about the expected input for certain fields, please refer to this example for guidance.

Bioinformatics Analysis

Is Bioinformatics Analysis required?

BIOINFORMATICS ANALYSIS REQUEST FORM

To provide the greatest value to our customers, Active Motif requires the following information be provided in conjunction with the sample submission form. This form is intended as a guide for our bioinformatics scientists to process samples using our standard pipelines in a way that is most compatible with the design and goals of each project. Any re-analysis that is required due to incomplete or inaccurate information may be charged at our standard bioinformatics rate. Active Motif may not be able to provide all requests on this form. For an overview of our standard offerings or deliverables, please see Active Motif Boinformatics Analysis . Contact your Account Manager for more information.

Data files will be available for 3 months after delivery, after which they will be deleted. Active Motif is not liable for data retention or retrieval after the deletion.

Provide a description of your experimental design.
Describe your goals for the bioinformatics analysis. (i.e., Identify Differential Regions)
To which species genome build do you want your data aligned? (Required*)
If you do not provide a specific request, Active Motif will use our default genome. (i.e., human: hg38, mouse: mm10, rat: rn6)
Bioinformatic service add-on: For an hourly rate we can perform data analysis and/or data integration with previous Active Motif Epigenetic Service data sets. If you would like this service add-on, please provide the quote number, sample names, and other relevant information.
Note: We may not be able to accommodate every request, and will reach out if necessary.
How should the samples be grouped for analysis? (i.e., S1: WT, S2: KO)
For ChIP-Seq, ATAC-Seq, RNA-Seq, RRBS, or CUT&Tag, how should the groups be compared for differential analysis? (i.e., WT vs KO)

(ChIP-Seq only) If we are generating input data from more than one sample, please specify which samples are to be analyzed with each input.
(Discover-Seq only) Please provide guide RNA (sgRNA) sequences for the samples to proceed with Discover-Seq analysis and identify double-stranded DNA cuts with respect to the CRISPR system.
If we are analyzing your data using an input from a previous Active Motif Epigenetics Service, please provide the project number and other specifics related to that input sample.
